A dose dependent response assay should be performed to determine the optimal concentration for use in specific applications. 1. ELISA and Western ELISA may be performed at dilutions of 1 : 2000 depending on the concentration of gp120 in the test system. CD4/gp120 capture ELISA may be performed in 1 : 500 dilution range depending on the capture ligand CD4 concentration and gp120 concentration in the test sample. Typical assays with 100ng of solid phase CD4 will detect at least 10ng/ml gp120.
